Dear Sir/Madam, I would like to help you out with your project, according to the PDF you have attached the PCB should be small in size, if you are planning to send to PCB to a manufacturer then the PCB layout design will be according to their design rules. if you are planning to make it manually then I will make sure the PCB is a single layer so that's it would be easier to fabricate. I have made similar PCBs before that also contained screw terminal components and standalone Arduino uno. I will make the design in Autodesk Eagle, which is an open source program so that you would be able to view the files without the need to buy any expensive software.
